Individuals have different behavioral traits that make up their personalities. Traits are
considered characteristics that both have a genetic basis and are shaped by experiences
throughout life and guide the way individuals behave in different situations and
environmental contexts experienced. The wide behavioral variability in a population is
important for species maintenance and individual survival. These characteristics may also be
related to risk behavior and influence the consumption, effects, and sensitivity of individuals
to psychoactive substances. Among them, alcohol and caffeine seem to be the most consumed
worldwide. Alcohol is the licit drug responsible for numerous diseases and accidents in
human beings, while caffeine, despite not having such serious effects, can also be harmful if
consumed in large quantities. Thus, the objective of this thesis was to evaluate the response of
behavioral profiles related to one of the personality traits, the bold-shy profile, under the
influence of alcohol and caffeine in zebrafish. In the first chapter, we assess the differences
between the profiles of a zebrafish population, to determine the risk-taking and exploration
behaviors that determine the two extremes of the bold-shy continuum. Then, we tested the
extreme profiles in two different contexts, an affiliative context (social group) and an aversive
context (presence of a predator), and under the effect of chronic and acute treatments regimen
with alcohol (chapter 2) and caffeine (chapter 3). Our results show that bold and shy profiles
show differences in risk-taking, bold individuals are more prone to risk than shy ones, and in
exploratory response, bold individuals are more explorative than shy ones. Furthermore, the
profiles respond differently to drug concentrations and analyzed contexts, with differences in
drug sensitivity, which makes bold individuals more sociable in some situations and shy
individuals more risk-averse in others. Our data show that there is a difference in
responsiveness between bold and shy, corroborating studies that relate personality to the effects of psychoactive substances. We hope these results will help advance further research
on the effects and treatments for substance abuse and its consequences. Finally, we reinforce
the use of zebrafish as an adequate and reliable tool for translational studies related to psychotropic drugs. |

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES Os indivíduos apresentam diferentes traços comportamentais que compõe sua personalidade. Os traços são considerados características que tanto tem base genética quanto são moldados pelas experiências ao longo da vida, e guiam o modo como os indivíduos se comportam nas diferentes situações e contextos ambientais vivenciados. A ampla variabilidade comportamental em uma população é importante para manutenção das espécies e sobrevivência dos indivíduos. Essas características podem também estar relacionadas a comportamento de risco e influenciar no consumo, efeitos e sensibilidade dos indivíduos à substancias psicoativas. Dentre elas, o álcool e a cafeína parecem ser as mais consumidas mundialmente. O álcool é a droga lícita responsável por inúmeras doenças e acidentes em seres humanos, enquanto a cafeína, apesar de não apresentar efeitos tão graves, também pode ser prejudicial se consumida em grandes quantidades. Dessa forma, o objetivo desta tese foi avaliar a resposta de perfis comportamentais relacionados a um dos traços de personalidade, o perfil bold-shy, sob efeito do álcool e da cafeína em zebrafish. No primeiro capítulo, avaliamos as diferenças entre os perfis de uma população de zebrafish, com o objetivo de determinar os comportamentos de tomada de risco e exploração que determinam os dois extremos do contínuo bold-shy. Em seguida, testamos os perfis extremos em dois contextos diferentes, um contexto afiliativo (grupo social) e um contexto aversivo (presença de predador), e sob efeito de tratamentos crônicos e agudos com álcool (capítulo 2) e cafeína (capítulo 3). Nossos resultados mostram que os perfis bold e shy apresentam diferenças na tomada de risco, indivíduos bold são mais propensos ao risco do que os shy, e na resposta exploratória, indivíduos bold são mais exploradores do que os shy. Ademais, os perfis respondem de forma distinta às concentrações de droga e aos contextos analisados, havendo diferenças na sensibilidade à droga, que torna indivíduos bold mais sociáveis em algumas situações e indivíduos shy mais avessos ao risco em outras. Nossos dados demonstram que há diferença na responsividade entre bold e shy, corroborando estudos que relacionam a personalidade aos efeitos de substâncias psicoativas. Esperamos que esses resultados ajudem no avanço de outras pesquisas sobre os efeitos e tratamentos para abuso de substâncias e suas consequências.
