Lipopeptides are biosurfactants, substances with a high surfactant potential
where each family corresponds to a group of isoforms that differs in the peptide's
composition and the length of the lipid chain. Among them, iturine and surfactin, lipopeptides
with high surfactant and antibiotic potential, stand out. As a lipopeptide, they are a set of
isoforms, biosynthesized or engineered, where the relationship between their structures and
properties helps choose and direct the product to the final application. That can range from
the surface cleaning and disinfection industry to crude oil recovery, in addition to being the
subject of studies in medical and pharmacological applications. In the context of COVID-19,
the present study aims to produce surfactin and iturin through the cultivation of Bacillus
subtilis UFPEDA 438, using sugarcane molasses as a substrate, as well as to evaluate his
biological activity in vitro, including his antiviral potential through in silico analysis of the
extract produced, against the 4 main target proteins of SARS-CoV 2. The results showed that
8 isoforms of surfactin and 2 of iturin were present in the samples, obtaining a maximum
productivity of 148.548 ± 4.672 mg/L of surfactin. The extracts produced in both steps
showed satisfactory levels and emulsification activities and promoted a reduction in the
surface tension of water by up to 55.24%. Against free radicals (ROS), the samples also
managed to block about 41.95% of them. Regarding the in silico evaluation, the greatest
interaction of the studied biosurfactants was with the Spyke protein, which is crucial for the
binding of the virus with the host human cell. The achieved results points to the extract
produced as a promising constituent in the elaboration of products with high antioxidant and
surfactant potential; and, in addition, moves future trials of his antiviral activity against SARS-CoV-2. |

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES Conselho Nacional de Desenvolvimento Científico e Tecnológico - CNPq Lipopeptídeos são biossurfactantes, substâncias com caráter tensoativo as quais cada família corresponde a um grupo de isoformas que diferem entre si na composição do peptídeo e do comprimento da cadeia lipídica. Dentre eles, se destacam a iturina e a surfactina, lipopeptídeos de alto potencial surfactante e antibiótico. Por se tratar de um lipopeptídeo, a surfactina é um conjunto de isoformas, biossintetizadas ou engenheiradas, na qual a relação entre suas estruturas e propriedades auxilia na escolha e no direcionamento do produto à aplicação final, indo desde a indústria de limpeza e desinfecção de superfícies à recuperação bruta de petróleo, além de ser alvo de estudos em aplicações médicas e farmacológicas. O presente estudo tem como objetivo produzir surfactina e iturina através do cultivo de Bacillus subtilis UFPEDA 438, bem como avaliar sua capacidade tensoativo, além do seu potencial antiviral através da análise in silico do extrato produzido frente as 4 principais proteínas alvo do SARS-CoV 2. Os resultados mostraram que 8 isoformas de surfactina e 2 de iturina estavam presentes nas amostras, obtendo-se uma produtividade máxima de 148,548 ± 4,672 mg/L de surfactina. Os extratos produzidos nas etapas 1, 2 e 3 da extração apresentaram índices e atividades de emulsificação satisfatórios e promoveram uma redução na tensão superficial da água em até 55,24 %. Frente aos radicais livres (ROS), as amostras também conseguiram bloquear cerca de 41,95% dos mesmos. Na avaliação in silico, a maior interação entre os biossurfactantes estudados foi com a proteína Spyke, que é crucial para a ligação do vírus com a célula humana hospedeira.
