It acts by inhibiting the formation of coagulation factors II, VII, IX and X, and therefore, an antagonist of vitamin K. In turn, drugs labeled with radioactive isotopes are commonly used in nuclear medicine for diagnostic purposes. As an example, there is technetium-99m, which is widely used radioisotope in human scans for diagnosis of various diseases and organ dysfunction. It has ideal physical characteristics for use in diagnostic nuclear medicine, such as is mono-gamma-emitting low energy (140keV), has a relatively short half-life (6.02 h) and emits no particulate radiation (α or β -). This study aimed to evaluate if the warfarin chronically administered for 10 consecutive days, alters the normal biodistribution of the radiopharmaceutical acid diethylene triamine pentaacetic labeled with technetium-99m (99mTc-DTPA) in the kidneys, bladder and blood of Wistar rat. For this, we used 12 Wistar rats, 3 months old and body weight ± 300 g, they were divided into control group (n = 6) and treated group (n = 6). The control group received 0.25 ml of filtered water and the treated group received 0.25 ml of warfarin solution (0,30mg/kg), both by gavage during 10 days. At the 10th day it was administered 0.1 ml of 99mTc-DTPA (0,66MBq radioactivity) via tail vein 1 hour after the treatment of two groups. After 1 h, all the animals were euthanasia for removal of the kidneys, bladder and blood sample. These samples were brought to the gamma counter to determine the percentage of the total injected activity per gram tissue (% ATI/g). There was a statistically significant increase in the % ATI/g of the kidney, respectively in the treated group compared to the control group (1.732±0.145 and 0.802±0.170) and a decrease in bladder (0.646±0,131 and 2.786±0.481 e) and blood (0.050±0.009 and 1.226±0.105) of treated group compared to control group, respectively. Based on these results, it appears that warfarin altered the normal 99mTc-DTPA biodistribution, probably, due to their biological and/or metabolites effects. |

Terapia com anticoagulantes orais como a varfarina é realizada na profilaxia de trombose e embolia em inúmeras desordens vasculares que afetam, em sua maioria, indivíduos com idade mais avançada. Ela atua inibindo a formação dos fatores de coagulação II, VII, IX e X, sendo, portanto, uma antagonista da vitamina K. Por sua vez, fármacos marcados com isótopos radioativos são muito utilizados na medicina nuclear para fins diagnósticos. A exemplo disso, tem-se o tecnécio-99m, que é o radioisótopo muito utilizado nas cintilografias humanas para diagnóstico de várias doenças e disfunções orgânicas. Ele apresenta características físicas ideais para a utilização em medicina nuclear diagnóstica, tais como: é mono-emissor gama de baixa energia (140keV), possui tempo de meia vida relativamente curto (6,02 h) e não emite radiação particulada (α ou β-). O presente estudo objetivou avaliar se o anticoagulante oral varfarina, administrado cronicamente por 10 dias consecutivos, altera a biodistribuição normal do radiofármaco ácido dietileno triamino pentacético marcado com tecnécio 99m (99mTc-DTPA) em rins, bexiga e sangue de ratos Wistar. Para tanto, utilizou-se 12 ratos Wistar com idade de 3 meses e massa corpórea ± 300 g, divididos em grupo controle (n=6) e grupo tratado (n=6). O grupo controle recebeu 0,25mL de água filtrada e o grupo tratado recebeu 0,25mL de varfarina (0,30mg/kg), ambos por gavagem e durante 10 dias. Ao 10º dia foi administrado 0,1mL de 99mTc-DTPA (0,66MBq de radioatividade), via veia caudal, 1 hora após o tratamento dos 2 grupos. Após 1 h, foi aplicada a anestesia e realizado eutanásia dos ratos com sucessiva retirada dos rins, bexiga e amostra de sangue. Essas amostras foram levadas ao contador de radiação gama para determinar o percentual de atividade total injetada por grama de tecido (%ATI/g). Observou-se um aumento estatisticamente significativo do %ATI/g nos rins, respectivamente do tratado em relação ao controle (1,732±0,145 e 0,802±0,170), e uma diminuição na bexiga (0,646±0,131 e 2,786±0,481) e no sangue (0,050±0,009 e 1,226±0,105) do tratado em relação ao controle, respectivamente.
