Among the CTCLs, the most common are mycosis fungoides (MF) with an indolent course and sezary syndrome (SS), which corresponds to the leukemic form of MF, generally associated with an aggressive course. The present study aimed to demonstrate the importance of immunophenotyping by flow cytometry through the positivity and intensity of antigenic expressions in confirming the diagnosis of mycosis fungoides and Sézary syndrome. Peripheral blood (SP) samples from 23 patients previously diagnosed with MF were analyzed in order to investigate SS. These patients were referred to the Laboratory of the Department of Hematology, Blood Center Dalton Barbosa Cunha (HEMONORTE), Natal, Brazil. Patients were analyzed for clinical, hematological and immunophenotypic aspects, using 4.0 mL of peripheral blood (SP). In immunophenotyping by flow cytometry (CF), specific panels of AcMo conjugated directly to three different fluorochromes were used: fluorescein isothiocyanate (FITC) Phicoerythrin (PE), chlorophyll pyridine protein, (PerCP), which detect green, orange and red fluorescence, respectively, allowing the labeling of up to three labels per tube, consisting of pan-B, pan-T and unspecified antigens. In our study, the CF analysis made it possible to identify the predominance of the population of mature T cells (CD3+/CD2+/CD5+, TCRAB+) with expansion of CD4+ cells in 100% of cases. Through Sézary cell (CS) analysis, patients numbers 1 and 3 were identified ( table 3 ) with absolute CS counts of less than 1,000/mm³, however, the other patients had absolute CS values > 1,000/mm³ of SP. In the results obtained, we observed that CF immunophenotyping proved to be an efficient methodology in the detection of CS cells, which can be used in the early diagnosis and monitoring of MS/SS.

Os linfomas cutâneos de células T (LCCTs) constituiem um grupo de doenças lifoproliferativas extranodais, atualmente classificadas e subdivididas de acordo com o comportamento clínico segundo a OMS/EORTC . Dentre os LCCTs, os mais comuns são a micose fungóide ( MF ) de curso indolente e síndrome sézary ( SS ), esta corresponde à forma leucêmica da MF, geralmente associada a um curso agressivo. O presente estudo teve como objetivo demonstrar a importância da Imunofenotipagem por citometria de fluxo por meio da positividade e intensidade de expressões antigênicas na confirmação do diagnóstico da micose fungóide e síndome de Sézary. Foram analisadas amostras de Sangue periférico (SP) de 23 pacientes previantente diagnosticados com MF no intuito de investigar a SS. Os referidos pacientes foram encaminados ao Laboratório do Departamento de Hematologia do Hemocentro Dalton Barbosa Cunha (HEMONORTE), Natal, Brasil. Os pacientes foram analisados quanto aos aspectos clínicos, hematológicos e imunofenotípico, utilizando 4,0 mL de sangue periférico (SP). Na Imunofenotipagem por citometria de fluxo (CF) foram utilizados painéis específicos de AcMo conjugado diretamente a três diferentes fluorocromos: isotiocianato de fluoresceína (FITC) Phicoeritrina (PE), proteína clorofila piridina, (PerCP), que detectam fluorescência verde, laranja e vermelha, respectivamente, possibilitando a marcação de até três marcações por tubo, constituidos de antígenos pan-B, pan-T e antígenos não espeficados. Em nosso estudo a análise por CF possibilitou identificar o predomínio da população de células T maduras (CD3+/CD2+/CD5+, TCRAB+) com expansão das células CD4+ em 100% dos casos. Mediante a análise de células de Sézary (CS) foram identificados os pacientes de números 1 e 3 ( tabela 3 ) com contagem absoluta de CS inferior a 1.000/mm³, no entanto, os demais pacientes apresentaram valores absolutos de CS > 1.000/mm³ de SP.
