Resumo: | The present work evaluated the influence of pH and sodium chloride using aqueous biphasic system (PEG–Phosphate) to lipase purification of Aspergillus niger produced by solid state fermentation using as substrate mangaba seed applying two planning with different molecular weight of Polyethylene glycol (PEG). The results showed the potential of mangabas’ seed flour as substrate and Aspergillus niger as microorganism to produce lipase by solid state fermentation. Experiment 2 in planning I, that is, to the system PEG 4000/ Phosphate (pH = 8,0, NaCℓ = 1,0%) was the one that presented the best results for partition coefficient (KP = 4,164), purification factor (FP = 1,629), and recuperation in upper phase (RS = 80,634%). Therefore, it can be concluded that the evaluated substrate presents potential for lipase production by solid state fermentation. |
