Hyaluronic acid (HA) is a high molar mass polysaccharide with countless applications in medical, pharmaceutical, esthetics and cosmetic products. Traditionally, this polymer is extracted from animal connective tissues such as umbilical cords and chicken combs. Therefore, the biotechnological production of HA, in particular Streptococcus zooepidemicus, appears as a promising alternative. In this context, the present study aims to produce, characterize and evaluate the in vitro antioxidant activity of hyaluronic acid produced by S. zooepidemicus CCT 7546. First, S. zooepidemicus strain was cultured by submerged fermentation in a rotary incubator at 150 rpm and 37 ° C for 22 hours. Then, the HA was extracted using sodium dodecyl sulfate (SDS) and purified by precipitation with 95% ethanol. Samples of HA produced by S. zooepidemicus and a commercial standard of sodium hyaluronate (SH) were subjected to Fourier transform infrared spectroscopy (FTIR), scanning electron microscopy (SEM), thermogravimetric analysis (TGA) and evaluation of antioxidant activity in vitro. The SEM analysis showed that SH presents itself as amorphous solids of uniform granulometry in a uniform medium, which corroborates the fact that it comes from an animal source, and the MEV of HA produced by S. zooepidemicus identified as a crystalline solid of granulometry non-uniform. In the FTIR curves characteristic bands were identified and typical to the two samples. Some organic functions have been identified as likely responsible for their antioxidant activity. In addition, activities were detected in the in vitro assays of DPPH radical sequestration, hydroxyl radical sequestration, reducing power and total antioxidant capacity. Finally, TGA results showed that AH of microbial origin is more thermally stable than its commercial equivalent of animal source. The results obtained also demonstrated the potential of AH obtained by biotechnology and contribute to the understanding of its characteristics and antioxidant activity. |

CNPq O ácido hialurônico (AH) é um polissacarídeo de alta massa molar com inúmeras aplicações em produtos médicos, farmacêuticos, estéticos e cosméticos. Tradicionalmente, este polímero é extraído de tecidos conjuntivos animais como cordões umbilicais e cristas de galo. Assim, a produção biotecnológica do AH, em particular por Streptococcus zooepidemicus, surge como alternativa promissora. Neste contexto, o presente trabalho tem como objetivo produzir, caracterizar e avaliar a atividade antioxidante in vitro do ácido hialurônico produzido por S. zooepidemicus CCT 7546. Inicialmente, cepa de S. zooepidemicus foi cultivada por fermentação submersa em incubador rotativo a 150 rpm e 37 °C por 22 horas. Em seguida, o AH foi extraído utilizando dodecil sulfato de sódio (SDS) e purificado por precipitação com etanol 95%. As amostras de AH produzido por S. zooepidemicus e um padrão comercial de hialuronato de sódio (HS) foram submetidas à caracterização por espectroscopia de infravermelho por transformada de Fourier (FTIR), microscopia eletrônica de varredura (MEV), análise termogravimétrica (TGA) e avaliação da atividade antioxidante in vitro. A análise de MEV mostrou que o HS se apresenta como sólidos amorfos de granulometria uniforme em um meio também uniforme o que corrobora com o fato de advir de uma fonte animal enquanto o MEV do AH produzido por S. zooepidemicus o identificou como um sólido cristalino de granulometria variável. Já a análise de FTIR identificou perfis característicos e comuns às duas amostras neles funções orgânicas foram identificadas como prováveis responsáveis por sua atividade antioxidante. Além disso, foram detectadas atividades nos ensaios in vitro de sequestro do radical DPPH, sequestro do radical hidroxila, poder redutor e capacidade antioxidante total. Por fim, os resultados de TGA mostraram que o AH de origem microbiana é mais estável termicamente que seu equivalente comercial de fonte animal.
