2-amino-4,5,6,7-tetrahydrobenzo[b]thiophene-3-carbonitrile (6CN), a 2-aminothiophene derivative, has potential pharmacological applicability. However, its
physical-chemical characteristics of 6CN impairs its clinical use. This work aimed to
develop binary systems between 6CN and β-cyclodextrin (βCD) in order to improve
the antiproliferative activity of 6CN in vitro, as well as to reduce its acute and
subchronic toxicity in vivo. Molecular modeling and phase solubility studies were
performed. The 6CN-βCD binary systems were prepared by physical mixture (PM),
kneading (KND) and rotaevaporation (ROTA). After the physical and chemical
characterization of binary systems, assays of antiproliferative activity in vitro were
performed on normal fibroblast cell lines (3T3), hepatic adenocarcinoma (HepG2)
and renal carcinoma (786-0) cells lines. Finally, acute and subchronic toxicity was
evaluated in vivo, using only the best system, to assess biochemical, hematological
and histopathological parameters. The characterization results show evident changes
in the physicochemical properties of 6CN after the formation of the binary systems with
βCD.Phase solubility studies indicated that βCD forms stable 1:1 complexes with 6CN,
corroborating with the molecular modeling data. The 6CN-βCD binary systems at
concentrations of 10 to 50µM, in the MTT assay, showed antiproliferative effects of
20–80% after 48h of exposure to 786-0 and HepG2 cancer cell lines, potentiating the
antiproliferative activity of 6CN. In vivo acute and subchronic toxicity tests were
conducted with ROTA, in which this binary system reduced the toxicity of 6CN, since
there was no mortality for animals treated with this system. The biochemical and
hematological findings did not show statistically significant changes indicative of
toxicity for animals treated with ROTA. In the histopathological analysis of acute
toxicity, hydropic degeneration was observed in the liver of animals treated with only
6CN. Therefore, the association between 6CN and βCD is a useful and promising
strategy to improve the physical and chemical characteristics of 6CN. Which May be
used in a future safe and effective anticancer therapeutic formulation, since the binary
systems increased the cytostatic effect and reduced the toxicity of 6CN. |

O 2-amino-4,5,6,7-tetrahidrobenzo[b]tiofeno-3-carbonitrila (6CN), um derivado 2- aminotiofeno, possui potencial aplicabilidade farmacológica, embora suas características físico-químicas dificultem seu uso. Com isso, o objetivo deste trabalho foi desenvolver sistemas binários entre o 6CN e a β-ciclodextrina (βCD) a fim de melhorar a atividade antiproliferativa in vitro do 6CN, bem como reduzir sua toxicidade aguda e subcrônica in vivo. Foram realizados estudos teóricos computacionais de modelagem molecular e diagrama de solubilidade de fases. Em seguida, foram desenvolvidos os sistemas binários entre o 6CN e a βCD pelos métodos de mistura física (MF), malaxagem (MAL) e rotaevaporação (ROTA). Após a caracterização físico-química dos sistemas binários, foram realizados ensaios de atividade antiproliferativa in vitro nas linhagens de fibroblastos normais (3T3), carcinoma hepático (HepG2) e carcinoma renal (786-0). E, por fim, avaliou-se a toxicidade aguda e subcrônica in vivo, apenas do melhor sistema, para avaliação dos parâmetros bioquímicos, hematológicos e histopatológicos. A caracterização físico-química mostrou que as características do 6CN foram alteradas após a formação dos sistemas binários entre 6CN e βCD. Os estudos de solubilidade de fase indicaram que a βCD forma sistemas estáveis na estequiometria de 1:1 com o 6CN, corroborando com os dados da modelagem molecular. Os sistemas binários em concentrações de 10 a 50 µM, no ensaio MTT, demostraram efeitos antiproliferativos de 20-80% após 48 horas de exposição a linhagens celulares cancerígenas 786-0 e HepG2, potencializando a atividade antiproliferativa do 6CN. Já os testes de toxicidade aguda e subcrônica in vivo foram conduzidos com o ROTA, em que esse sistema binário reduziu a toxicidade do 6CN, uma vez que não houve mortalidade para os animais tratados com esse sistema. Os achados bioquímicos e hematológicos não demonstraram alterações estatisticamente significativas indicativas de toxicidade para os animais tratados com o ROTA. Já na análise histopatológica da toxicidade aguda observou-se degeneração hidrópica no fígado dos animais tratados apenas com o 6CN.
