Laminarin (1,3-β-glucan) and fucan (sulfated polysaccharide consisting of sulfated Lfucose) are synthesized by brown seaweeds and have several pharmacological
activities described. However, polysaccharides and their activities can be modified,
increasing their activities. Thereunto, various techniques are used, such as those
called green methods. Given the above, and bearing in mind that the potiguar coast,
as well as the northeastern, holds several species of seaweed that have not had their
neutral and sulfated polysaccharides completely studied, this work aimed to prospect
the main chemical modifications in neutral polysaccharides (laminarins) from
Lobophora variegata and sulfated polyscarides (fucanas) from Dicytiota mertensii
and verify their farmacological activities, choosing the best modifications for structural
characterization. Algae were collected at Pirangi beach, Natal, RN, washed and
submitted to proteolysis. Using acetone differential fractionation and molecular mass
separation techniques, a 12.4 kDa laminarin was obtained. Physicochemical analysis
showed that laminarin (LM) consists only of glucose. Dielectric barrier discharge
(DBD), sulfation and galification techniques were used to obtain modified laminarins
called LMC, LMS and LMG. The modifications promoted different amounts functional
groups inserted: for LMS 1.4% of sulfate was observed, for LMG 1.5% of gallic acid
and LMC 11.7% of uronic acids. The modification that most enhanced the antioxidant
activities of laminarins was galification, therefore, LM and LMG had their structure
elucidated by Nuclear Magnetic Resonance (NMR). The data obtained by NMR
analysis corroborate with the previous data proving the structure of laminarin in LM
and gallified laminarin in LMG. LM and LMG were not cytotoxic and did not alter the
cell cycle of normal canine kidney cells (MDCK) compared to the control. In parallel, it
was possible to obtain fucans from D. mertensii after proteolysis and precipitation
with methanol. Therefore, silver nanoparticles with fucan called NF were synthesized
and called NF. The seventh day was the optimal synthesis day for this particle,
whose size was approximately 104 nm. The NF had a round shape very
homogeneous size distribution, and stability for 16 months and showed a higher
antitumor activity (1mg / mL) compared to fucan. NF also had better antibacterial
activity than native fucan and immunomodulatory activities (nitric oxide production
and cytokine production) had similar results when compared to NF with native
fucans. In general, it we observed that the gallification was the best chemical modification in L. variegata laminarins and the NF potentiated the pharmacological
activities of the fucans. |

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) Laminarinas (1,3-β-glucanas) e fucanas (polissacarídeos sulfatados constituídos de L-fucose sulfatada) são sintetizadas por algas marrons e possuem diversas atividades farmacológicas descritas. Todavia, os polissacarídeos e suas atividades podem ser modificados, potencializando-as ou não. E para tal, são utilizadas várias técnicas, como por exemplo, aquelas denominadas de métodos verdes. Diante do exposto, e tendo em mente que o litoral potiguar, assim como o nordestino, detém várias espécies de algas marinhas que não tiveram seus polissacarídeos neutros e sulfatados estudados completamente, este trabalho teve como objetivo realizar uma prospecção das principais modificações químicas em polissacarídeos neutros (laminarinas) da alga Lobophora variegata e polisscarídeos sulfatados (fucanas) da alga Dicytiota mertensii e verificar as suas atividades farmacológicas, escolhendo as melhores modificações para caracterização estrutural. As algas foram coletadas na praia de Pirangi, Natal, RN, lavadas e submetidas à proteólise. Com o uso de fracionamento diferencial com acetona e técnicas de separação por massa molecular, foi obtido uma laminaria de 12,4 kDa. Análises físico-químicas mostraram que a laminarina (LM) era constituída apenas de glicose. As técnicas de descarga da barreira dielétrica (DBD), sulfatação e galificação foram utilizadas paar se obter as laminarinas modificadas denominadas LMC, LMS e LMG. As modificações promoveram diferentes quantidades de inserção dos grupos funcionais: com LMS observou-se 1,4% de sulfato, com LMG observouse 1,5% de ácido gálico e com LMC observou-se 11,7% de ácidos urônicos. A modificação que mais potencializou as atividades antioxidantes de laminarinas foi a galificação, por isso, LM e LMG tiveram sua estrutura elucidada por Ressonância Magnética Nuclear (RMN). Os dados obtidos pelas análises de RMN corroboram com os dados anteriores, comprovando a estrutura da laminarina em LM e laminarina galificada em LMG. LM e LMG não foram citotóxicas e não alteraram o ciclo celular de células normais de rim canino (MDCK) em comparação com o controle. Em parelelo, foi possível obter fucanas da alga D mertensii por meio de proteólise e precipitação com metanol. Além disso, sintetizou-se nanopartículas de prata com a fucana, que foi denominada de NF. O sétimo dia foi o dia ótimo de síntese desta partícula, cujo tamanho foi de aproximadamente 104 nm. As NF tiveram forma arredondada e distribuição do tamanho bastante homogêneo, estabilidade por 16 meses e apresentaram uma maior atividade antitumoral (1mg/mL) comparada à fucana. Com NF também se obteve melhor atividade antibacteriana do que com a fucana nativa e as atividades imunomodulatórias (produção de oxido nítrico e produção de citocinas) tiveram resultados semelhantes comparando-se NF com fucanas nativas.
