In this work, we evaluated the healing effects of a new zinc complex associated with a chitosan membrane, tested in cutaneous wounds of diabetic rats. Wistar rats were submitted to induction of diabetes with streptozotocin 40 mg / kg (i.v.). On the seventh day after diabetic induction, a circular skin excision (1.0 cm) with punch was performed. The lesions were treated with the pure chitosan membrane and with the membrane associated with the Zinc-vanillin complex (CS-ZPV1 and CS-ZPV2) at two concentrations, 7.36 x 10-7 mol cm-2 and 1.22 x 10-6 mol cm -2, respectively. The samples were submitted to macroscopic, histopathological, cytokine (TNFα, IL-1β and IL-10) and reverse transcriptase polymerase chain reaction (RtPCR) (TGFβ and VEGF) assays. The analysis revealed reepithelialization, angiogenic stimulation, collagen deposition and expression of proteins that aid in the healing process in a significant proportion in the groups treated with the CS-ZPV membrane compared to the groups without treatment. Treatment with CS-ZPV reduced the levels of TNF-α, IL-1β and increased IL-10, also increased the gene expression of TGFβ and VEGF. The evaluated parameters suggest that CSZPV in the two concentrations tested may be effective in the treatment of chronic wounds. Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) O tratamento de feridas crônicas ainda é considerado um problema de saúde pública, quando afeta grupos de riscos como diabéticos e torna-se um grande desafio clínico. Neste trabalho, avaliamos os efeitos cicatrizantes de um novo complexo de zinco associado a uma membrana de quitosana, testado em feridas cutâneas de ratos diabéticos. Ratos do tipo Wistar foram submetidos a indução do diabetes com estreptozotocina 40 mg/kg (i.v.). No sétimo dia após a indução diabética, foi realizada uma excisão circular na pele (1,0 cm) com punch. As lesões foram tratadas com a membrana de quitosana pura e com a membrana associada ao complexo de Zinco-vanilina (CS-ZPV1 e CS-ZPV2) em duas concentrações 7.36 x 10-7 mol cm-2 e 1.22 x 10-6 mol cm-2 , respectivamente. As amostras foram submetidas à análise macroscópica, histopatológica, dosagens de citocinas (TNF-α, IL-1β e IL-10) e reações em cadeia de polimerase de transcriptase reversa (RtPCR) (TGFβ e VEGF). As análises realizadas demonstraram reepitelização, estímulo angiogênico, deposição de colágeno e expressão de proteínas que auxiliam no processo de cicatrização em proporção significativa nos grupos tratados com a membrana CS-ZPV comparado aos grupos sem tratamento. O tratamento com a CS-ZPV reduziu os níveis de TNF-α, IL-1β e aumentou IL-10, verificou-se também o aumento da expressão gênica de TGFβ e VEGF.
