Drugs are the group of emerging contaminants that have been receiving attention in recent
years, with hospital effluents being the predominant source of introduction into the
environment. The main concern related to the presence of these compounds in the
environment is due to the development of bacterial resistance. The antibiotics stand out
because of their intensive use, and it can be emphasized those that fight Tuberculosis
considered one of the biggest problems of health worldwide. In view of this conjuncture, it
was proposed to develop and validate a liquid chromatography metodology coupled to tandem
mass spectrometry with electrospray ionization for the analysis of anti-tuberculosis drugs via
direct injection in hospital effluent samples. The developed method complied with all required
validation parameters. The method robustness was evaluated through the Youden test, which
allowed to determine that the best chromatographic condition was the one using mobile phase
flow of 0.150 mL min
-1
, column temperature of 25°C, without sample acidification and using
Acclain Polar Advanced (C16). The method presented good linearity in the working range of
0.1 to 100 μg L
-1 with R2
values higher than 0.9632 for the four drugs. The method was
shown to be sensitive, accurate, robust and with recoveries in accordance with INMETRO,
above 80%. LDs and LQs were respectively 0.028 and 0.057 μg L
-1
for Pyrazinamide, 0.094
and 0.176 μg L
-1
for Isoniazid, 0.416 and 0.833 μg L
-1
for Rifampicin and 0.263 and 0.519 μg
L
-1
for Ethambutol. Regarding Hospital Effluent samples, no Ethambutol was detected in any
samples, while Isoniazid was determined in all samples at concentrations between 3.25 and
23.09 μg L
-1
. Therefore, it can be concluded that the developed method can be applied for
determination of these pharmaceutical compounds with high precision and sensitivity. |

Os fármacos constituem o grupo de contaminantes emergentes que vem recebendo a atenção nos últimos anos, sendo os efluentes hospitalares a fonte predominante de introdução desses no ambiente. A principal preocupação relacionada à presença desses compostos no meio ambiente é devido ao desenvolvimento de resistência bacteriana. Os antibióticos se destacam por causa do seu uso intensivo, podendo-se enfatizar os que combatem à Tuberculose considerada um dos maiores problemas de saúde mundial. Diante dessa conjuntura, foi proposto desenvolver e validar uma metodologia de cromatografia líquida acoplada à espectrometria de massas em tandem com ionização por eletrospray, para a análise de fármacos anti-tuberculose via injeção direta em amostras de efluente hospitalar. O método desenvolvido cumpriu com todos os parâmetros de validação exigidos. A Robustez do método foi avaliada através do teste de Youden, que permitiu determinar que a melhor condição cromatográfica foi a que utiliza a vazão da fase móvel de 0,150 mL min-1 , temperatura da coluna de 25°C, sem acidificação da amostra e utilizando a coluna Acclain Polar Advanced (C16). O método apresentou boa linearidade na faixa de trabalho de 0,1 a 100 µg L -1 com valores de R2 superiores 0,9632 para os quatro fármacos. O método demonstrou-se sensível, preciso, robusto e com recuperações em conformidade com o estabelecido pelo INMETRO, superiores a 80%. Os LDs e os LQs foram respectivamente de 0,028 e 0,057 µg L -1 para a Pirazinamida, de 0,094 e 0,176 µg L -1 para a Isoniazida, de 0,416 e 0,833 µg L -1 para a Rifampicina e de 0,263 e 0,519 µg L -1 para o Etambutol. Em relação as amostras de Efluente Hospitalar, não foi detectado Etambutol em nenhuma das amostras, enquanto a Isoniazida foi determinada em todas as amostras, nas concentrações entre 3,25 e 23,09 µg L -1 .
