Resumo:ABSTRACT Brown algae present polysaccharides of biological interest. Herein, we describe the isolation and characterization of a fucan and laminarin isolated from acetone fractionation. This fraction was further fractioned by gel filtration chromatography yielding two additional peaks containing a high molecular weight compound, a galactofucan of 1,400 kDa and a laminaran of 11 kDa. The galactofucan presents a molar ratio of fucose, galactose and sulfate, 1:3:2, respectively. Infrared spectrum analysis of this compound showed a small peak at 1.734 cm ¯¹ that may be assigned to ester carbonyl resonances, thus confirming the presence of acetate in this galactofucan, whereas the absorption band at 851 cm ¯¹ corresponded to the axial sulfate. ¹³C nmr spectrum showed a signal at 16.5 ppm, related to methyl group of a-fucopyranosides. The signals at 19-20 ppm confirmed the presence of acetyl group, previously shown in other studies. Several signals in the anomeric region at 93 ppm could be assigned to α-L-fucose residues and 104 ppm was assigned to β-d-galactose residues. The laminarin is composed only of glucose and the IR spectrum revealed the absorption band at 889 cm ¯¹, usually seen in β-glucans. In the anomeric region at least two signals were observed. HMQC suggests a compound without side chains in C6. Signals at 62 ppm is common for C6 to the 1->3 linked residues. The citotoxicity of those compounds were analyzed using HL60 cells treated with L4, subtractions of the galactofucan (GFL1-1.600 kDa, GFL2-1.200 kDa and GFL-390 kDa), laminaran and commercial fucoidan. Parameters on mitochondrial dehydrogenase, content of proteins and fosfatase activity were altered. Further investigations are needed to exploit the probable biological and pharmacological roles of those polysaccharides. Key words: Brown algae, fucans, glucans, HL60 cells.
